The workshop is meant to cover recent advances in melanoma research, from basic science to clinical applications. The workshop will provide the frame of a focused panel of discussion covering all the topical aspects of transcriptional regulation, molecular pathways and novel therapeutic approaches in melanoma progression, through the study of single molecules, cell-host interactions and model organisms.
The workshop will include broad lectures and invited talks directed at graduate students, postdoctoral fellows and researchers. Invited lecturers will present questions, approaches and results from their ongoing research in the field.

The aim of the conference is to foster a deep exchange among leading experts on the theme of epigenetic reprogramming, drawing fresh insight from the juxtaposition of the main lines of inquiry that are shaping this field. Lineage choices, their stability and their reversibility, are now at the crossroad of several research fields which range from the basic biology of differentiation and morphogenesis to the biotechnological engineering that is reshaping lineages for medical applications. Nuclear transfer, cell fusion and induced pluripotency have defined the past decade with radical challenges to many long standing tenets of developmental biology and have propelled a new paradigm for regenerative medicine. Meanwhile advances in lineage tracing for the main model organisms have uncovered salient examples of in vivo reprogramming, either spontaneous or induced by genetic or epigenetic manipulations. We believe the time is ripe to take stock of these converging trajectories and dedicate an ad hoc effort to map the current state of reprogramming as a research field in its own right, with the depth and breadth of analysis that can only come from the interdisciplinary contribution of the various strands of ‘reprogramming’ research. Hence this meeting attempts to carve an intellectual space distinct from the format of either all-encompassing stem cell conferences or highly specialiazed workshops devoted to a single cell or signaling pathway. At this defining time in its growth, the intellectual venture of reprogramming needs both focus and lateral thinking.

The horizon for cancer immunotherapy has been widened by recent acquisitions on the biology of dendritic cells and by research on the role of T regulatory cells in cancer immune escape. Brain tumors are an excellent arena for the development of this progress. Malignant gliomas, in particular, are the object of novel trials that have as a background the initial experience gained with dendritic cell immunotherapy. Moreover, new insights on the role of cancer stem-like cells in glioma development may help defining more specific targets for clinically relevant immune responses.

Understanding how cells migrate contributes to elucidate various fundamental processes involved in normal and pathological conditions. A significant progress in unraveling the mechanisms underlying cell migration can only be achieved through a comprehensive approach spanning from the study of single molecules to model organisms and diseases. The course is aimed at elucidating some of the molecular pathways involved in the progression of normal melanocytes to melanoma cells. The course will have a combination of teaching lectures, research talks and practicals.
The first part will deal with the analysis of the structural characteristics of the melanocytes, their phenotypic changes when melanoma develops, therapeutic overview and an update on the current genetic manipulation methodologies, which might be applied to these cells under specific growth conditions.
The second part will focus on the use of novel technologies for melanocyte and melanoma analysis and on the discussion of mouse models in the context of development and migration.
Practical sections will be devoted to: 1) the definition of skin cell types and the analysis of the phenotypic characteristics of malignancy; 2) the acquisition of the protocols of melanocyte and melanoma processing and culture and 3) the preparation of tissue microarrays from tumoral biopsis.
This is a course for graduate students, postdoctoral fellows, and young researchers willing to translate knowledge of the biology of melanocytic cells to a practical approach in the field. Invited lecturers will present questions, approaches and results from their ongoing research in the field.
The course will be kept small, with a maximum of 40 participants. Preference will be given to active participants selected on the basis of their submitted curricula.

The workshop was composed of a theoretical and a practical part. In the theoretical part experts on dendritic cell-based vaccinations were invited to report on their experience in cancer immunotherapy clinical trials, including failures and successes. In the practical part, selected applicants participated to two laboratory workshops where protocols on how to derive dendritic cells from blood precursors and how to monitor the induction of immune responses in patients were showed and discussed.
